Can the variables from EMWPROF.EXE be used with the Mail Source Agent Switch Message (4228076)

Variables in switch message are not showing anything but the variable (i.e. %U instead of Username)

This is by design

The messages for the actual mailbox switch pulls the "Target Server Name" and "Target Mailbox DN" from the SQL database as part of the switch process.   The text of the message can be changed by editing the switch message, however; outside variables, such as those used in EMWPROF.EXE have no effect on the message.   You can insert the "Target Server Name" and "Target Mailbox DN" variables in your edited message by clicking the hyperlink on the dialog box.
