If 1 virtual user is set in the general setting page, why did the number of virtual user increased during the time when the Relay Trace job is running?

Cause
Benchmark Factory setting page is a general settings for default values. The default values will be overridden if the test contains its own settings for these values.

Resolution
In Replay Trace test, the number of virtual user is controlled by the number of sessions in the trace file.
Each session in the trace file gets assigned to single virtual user who executes only that session. -

This applies to both SQL Server and Oracle Trace.
